Taxonomic characteristics and protease activity exploration of marine heterotrophic bacteria isolated from the coast of Eulwangni in the West Sea, Korea

Abstract
Heterotrophic bacteria play an important role in the cycling of organic matter in marine environments, and the proteases they produce have high commercial values, therefore more research into them is required. Herein, we isolated and cultured heterotrophic bacteria from two different environments (sea water and mud flats) at Eulwangni Beach in Incheon, Korea.
Their taxonomic identities were determined by using the 16S rDNA sequences, and they were further evaluated for protease productivity according to the size of the clear zone. We successfully isolated a total of 14 species from 54 isolates from St. 1 (sea water) and St.2 (mudflat). Based on the 16S rDNA sequence comparisons, they taxonomically belonged to Actinobacteria, Firmicutes, and Proteobacteria. The most common taxa found at both sampling sites were Proteobacteria. However, the dominant taxa were different: St. 1 was recorded as Actinobacteria, while St.2 was Firmicutes. We discovered high protease activity in 28 isolates, from Bacillus altitudinis, B. stratosphericus, B.
toyonensis, B. xiamenensis, Micrococcus luteus, and Rheinheimera aquimaris. These results revealed the taxonomic features and protease production characteristics of heterotrophic bacteria isolated from Eulwangni, Incheon, suggesting the possibility of future microbial resource utilization.
